CÅSDROP

CasDrop is an optogenetic inducible condensate module similar to the Corelet system. While in the latter sticky IDRs get recruited to a preformed ferritin core, in CasDrop multiple IDRs target a synthetic tail with tandem binding sites that is fused to Cas9. Co-expression with a guide RNA allows the specific formation of condensates at a precise genomic position.

Other optogenetic condensate systems are optoDroplets, Corelets and PixELLs.

PROS:
+ common molecular grammar of IDR
+ IDR subunit is highly tunable
+ precise genomic targeting
+ optogenetic control

CONS:
- large constructs
- potential interference with endogenous processes depending on IDR
